I know it's intended, but I want to know if it's a glitch or not. I want to speedrun any% glitchless no S + Q and I want confirmation if collecting this ruppee is legal or not in a glitchless run?

It's not a glitch since it's the way Nintendo intended for the player to get that rupee, and as Nintendo isn't intending for the intended way to get something to be via a glitch, I'm sure that it is speedrun-legal. Happy running!
